چکیده

Background Stimulators of the nitric oxide (NO) receptor soluble guanylate cyclase (sGC) are a new class of substances which enhance the biosynthesis of cyclic guanosin monophosphate (cGMP) independently from the presence of nitric oxide (NO). In pulmonary smooth muscle cells, increased intracellular cGMP concentrations lead to vasodilatation, making sGC-stimulators potentially useful for the treatment of patients suffering from pulmonary hypertension (PH). Despite recent advances in the management of PH there is still significant morbidity and mortality from this condition which may occur in isolation (idiopathic pulmonary arterial hypertension, iPAH) or in the context of different clinical settings including chronic thromboembolic pulmonary hypertension (CTEPH), PH in chronic obstructive pulmonary diseases (PH-COPD), in interstitial lung disease (PH-ILD) or in left heart disease (PH-LHD).

Renal effects of soluble guanylate cyclase stimulators and activators: a review of the preclinical evidence.

Direct stimulation of soluble guanylate cyclase (sGC) is emerging as a potential new approach for the treatment of renal disorders. sGC catalyzes the formation of cyclic guanosine monophosphate (cGMP), deficiency of which is implicated in the pathogenesis of chronic kidney disease (CKD). Therefore, new classes of drugs - sGC stimulators and activators - are being investigated in preclinical mod...

Modulation of soluble guanylate cyclase for the treatment of erectile dysfunction.

Nitric oxide (NO) is the principal mediator of penile erection, and PDE-5 inhibitors are the first-line agents used to treat erectile dysfunction (ED). When NO formation or bioavailability is decreased by oxidative stress and PDE-5 inhibitors are no longer effective, a new class of agents called soluble guanylate cyclase (sGC) stimulators like BAY 41-8543 will induce erection. sGC stimulators b...

New insights into the role of soluble guanylate cyclase in blood pressure regulation.

PURPOSE OF REVIEW Nitric oxide (NO)-soluble guanylate cyclase (sGC)-dependent signaling mechanisms have a profound effect on the regulation of blood pressure (BP). In this review, we will discuss recent findings in the field that support the importance of sGC in the development of hypertension. RECENT FINDINGS The importance of sGC in BP regulation was highlighted by studies using genetically...

Stimulators and Activators of Soluble Guanylate Cyclase: Review and Potential Therapeutic Indications

The heme-protein soluble guanylyl cyclase (sGC) is the intracellular receptor for nitric oxide (NO). sGC is a heterodimeric enzyme with α and β subunits and contains a heme moiety essential for binding of NO and activation of the enzyme. Stimulation of sGC mediates physiologic responses including smooth muscle relaxation, inhibition of inflammation, and thrombosis. In pathophysiologic states, N...

Additional stimulation of sGC on top of standard treatment with ARB`s may offer a new therapeutic approach for the treatment of diabetic nephropathy resistant to ARB treatment alone

Background Riociguat is the first of a new class of drugs, the soluble guanylate cyclase (sGC) stimulators. Riociguat has a dual mode of action: it sensitizes sGC to the body’s own NO and can also increase sGC activity in the absence of NO. The NO-sGC-pathway is impaired in many cardiovascular diseases such as heart failure, pulmonary hypertension and diabetic nephropathy (DN). DN leads to high...
